Validation of the Lumi-cell Er Recombinant Bioassay for Rapid Evaluation of Chemicals for Potential Estrogenic Activity

چکیده
Introduction The association between the exposure and bioaccumulation of endocrine disruptor chemicals (EDCs) and their adverse effects on human and wild life populations has raised concern worldwide. Due to the detrimental effects of environmental exposure to EDCs, there is an obvious need to develop a relevant bioassay, which can both detect these chemicals, as well as provide a relevant estimate of their endocrine disrupting potency. Some examples of the effects of EDCs are: decreased reproductive success and feminization of males in several wildlife species; increased hypospadias along with reductions in sperm counts in men; increase in the incidence of human breast and prostate cancers; and endometriosis . Because these chemicals are ubiquitous, highly lipophilic, and often chlorinated, ensures their persistent presence in the environment resulting in their bioaccumulation in the food chain.

Bioassay Methods useful to select the natural product cancer chemopreventive agents
Prevention of malignant disease such as cancer is obviously much more important than therapy, so primary prevention tactics should be invoked with the greatest possible effort. Because delaying a disease such as cancer is somehow equivalent to cure and it is now generally accepted that cancer can be managed through chemopreventive interventions. Since a considerable number of known cancer chemo...

متن کاملEstrogenic Activity of Some Phytoestrogens on Bovine Oxytocin and Thymidine Kinase-ERE Promoter through Estrogen Receptor-α in MDA-MB 231 Cells
Background: Phytoestrogens, a group of plant-derived polyphenolic compounds have recently come into considerable attention due to the increasing information on their potential adverse effects in human health. Some of phytoestrogens show estrogenic activity that may be carcinogenic for human. In the present study, we investigated the transcriptional effects of variety of phytoestrogens ...
متن کاملAntimutagenic, antitumor and estrogen receptor binding activity of the rare plant Shortia galacifolia: An ethnobotanical and chemosystematic approach
Objective: Shortia and other members of the Diapensiaceae family have ethnomedicinal history in both Eastern and Western hemispheres. Based on ethnopharmacological and chemosystematic evidence, pharmacological and toxicological bioassays were conducted on the rare plant Oconee Bell, Shortia galacifolia. Materials and Methods: Extracts were examined in assays for antimutagenicity, antitumor and ...
متن کاملMeasurement of estrogenic activity in sediments from Haihe and Dagu River, China.
Sediments from two rivers in China, the Haihe and Dagu Rivers, were examined for estrogenic activity using an estrogen receptor (ER)-mediated in vitro bioassay system. ER-active compounds were isolated from sediments by Soxhlet extraction, and the crude extracts were fractionated using a florisil column into three fractions. The estrogenic activity of each extract was detected by measuring luci...
